Diabetic retinopathy is a severe complication of diabetes mellitus. Results of this study showed that intravitreal injection of mesenchymal stem cell-derived exosomes could inhibit microglial activation and alleviate diabetic retinopathy. https://doi.org/10.1093/stmcls/sxad078

Understanding the detailed molecular mechanisms of embryogenesis is necessary for the development of embryology and regenerative medicine. This study identified a crucial metabolic pathway to regulate the formation of primitive streak.

Abstract. In early embryogenesis, the primitive streak (PrS) generates the mesendoderm and is essential for organogenesis. However, because the PrS is a minute

The paucity of biomarkers that predict potency prior to patient administration limits the efficacy of MSC-based therapy. This article reports the use of TWIST1 as a biomarker of MSC potency.

Abstract. Despite extensive clinical testing, mesenchymal stem/stromal cell (MSC)-based therapies continue to underperform with respect to efficacy, which refle

Regenerative treatment of damaged cartilage, especially for osteoarthritis, is a promising treatment, but obtaining chondrocytes in sufficient numbers is a limiting factor. This report of a molecular adjustment to SOX9 structure leads to improvement of the current method of chondrocyte generation by direct reprogramming. The results showing that the novel SOX9 variant may be useful for efficient induction of chondrocytes points to the strategic feasibility of mutating a transcription factor at functionally critical residues to optimize reprogramming. https://doi.org/10.1093/stmcls/sxad066

It was believed that lower expression in Pax6 is the main factor for the observed limbal stem cell deficiency-like phenotype in patients with aniridia. This study showed this is not the case, pointing to the role of Pax6 in the niche as the main underlying factor for the disease.

Abstract. Congenital aniridia is caused by heterozygous mutations on the PAX6 gene leading to reduced amount of PAX6 protein (haploinsufficiency), abnormal eye

Selective degeneration of retinal ganglion cells (RGCs) in glaucoma leads to irreversible vision loss. Results of this study suggest that ex vivo generated human RGCs may be a suitable stem cell approach to treat RGC degeneration glaucoma.

Abstract. Retinal ganglion cells (RGCs) connect the retina with the higher centers in the brain for visual perception. Their degeneration leads to irreversible

This article reports the use of autologous iPSC-based photoreceptor cell replacement to restore vision in patients with inherited retinal degenerative blindness.

Abstract. Inherited retinal degeneration is a term used to describe heritable disorders that result from the death of light sensing photoreceptor cells. Althoug

This work examined bone marrow cell types through single-cell RNAseq to define new roles for signal transducer and activator of transcription-5 ( ). Results showed that STAT5ab-associated gene regulation supports the bone marrow microenvironment.

Abstract. Signal transducer and activator of transcription 5 (STAT5a and STAT5b) are intrinsically critical for normal hematopoiesis but are also expressed in s
